Principle of the Test:
In this assay the Fibrinogen present in samples reacts with the anti-Fibrinogen antibodies which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, anti-FIB antibodies conjugated with horseradish peroxidase (HRP), are added. These enzyme-labeled antibodies form complexes with the previously bound FIB. Following another washing step, the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate, 3,3?,5,5?-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of FIB in the sample tested; thus, the absorbance, at 450nm, is a measure of the concentration of FIB in the test sample. The quantity of FIB in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.

Fibrinogen, Human, BioAssay ELISA Kit is a highly sensitive two-site enzyme linked immunoassay (ELISA) for measuring Fibrinogen in biological samples of humans.



Kit Components

F4202-02H1: Diluent Concentrate (Running Buffer): 1x 50ml of a 5X concentrated diluent running buffer.
F4202-02H2: Wash Solution Concentrate: 1x50ml of a 20X concentrated wash solution.
F4202-02H3: Enzyme-Antibody Conjugate 100X: 1x150ul of affinity purified anti-Human Fibrinogen antibody conjugated with horseradish peroxidase in a stabilizing buffer.
F4202-02H4: Chromogen-Substrate Solution: One vial containing 1x12ml of 3,3?,5,5?-tetramethybenzidine (TMB) and hydrogen peroxide in citric acid buffer at pH 3.3.
F4202-02H5: Stop Solution, 0.3M sulfuric acid: 1x12ml
F4202-02H6: Microtiter Strip: 1x96wells in well holder frame. Each well is coated with affinity purified anti-Human FIB.
F4202-02H7: Human Fibrinogen Calibrator: 1×1 vial lyophilized Human Fibrinogen calibrator.

Storage and Stability

Store *F4202-02H7 powder at 4°C. Once reconstituted, aliquot and store at -20°C. Store all other components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
